Detection of succinic acid in vitamin B12-enriched tempeh by HPLC

High perfomance liquid chromatography (HPLC), Zorbax ODS column detected at the wavelength of 220 mm was used to determine the succinic acid in vitamin B12-enriched tempeh, a fermented soybean produced by mixed fermentation of the mold Rhizopus oligosporus and the bacteria Propionibacterium shermanii 1250. The mixture of 1.5 % KH2PO4 and ACN at the ratio of 95:5 was used as mobile phase at the flow rate 1 ml/min. Increase of succinic acid was observed in the tempeh at 24 hr and then, decreased at 48 hr of fermentation time. It can be seen that the change in succinate in the tempeh may be directly related to the formation of vitamin B12 in tempeh by the bacteria P. shermanii 1250.
