Effect of esterases inhibitors on gamma-irradiated Culex pipiens L. females

Inhibitory effect of eserines, dichlorovos, azodrin and paraoxone on non-irrdiated and irradiated adult C. pipiens female esterases was examined. Two esterase bands (Rm 0.37 and 0.45) were activated or digested alpha-naphthyl acetate by the addition of the two inhibitors azodrin and paraoxone respectively, which were not detected on gel before the inhibitors were added to the substrate. Carboxyesterase and cholinesterase isozymes were mostly detected, whereas arylesterase was rarely detected. Nine carboxyesterase bands were detected in A1 phase increased by increasing the age to ten bands at A3 and A7. Cholinesterase enzyme was detected in five bands at A1 phase increased to six bands at A3 and decreased to four at A7. Exposure to gamma irradiation at sub-sterilizing and sterilizing dose levels resulted in a fluctuating effect on both carboxyesterase and cholinesterase, however this effect was completely disregarded in the case of arylesterase. After exposure to gamma radiation, the number of detected carboxyesterase bands was decreased at day 7 from ten to seven bands. While the number of cholinesterase bands increased at day 1 to six bands after exposure to 70 and 80 Gy, then decreased to four and three bands at day 3 and 7. The present investigation demonstrated that the rate of enzyme activity was not proportional to the applied dose. The effectiveness of gamma-radiation on the esterase isozymes may be the result of difference in sensitivity of the esterase loci which control biosynthesis of the enzyme active protein
