Separation of esterase and peroxidase isozymes in plants by disc gel electrophoresis

The Disc Polyacrylamide Gel Electrophoresis (disc PAGE) was used in this study for determination of plant isozymes. As being a genetic marker, esterase and peroxidase isozymes in crude extract of various kinds of plant such as mango, papaya, mungbean, soybean, rice, sweet sorghum, corn, wheat and sugarcane, were separated by disc PAGE with appropriate conditions. The isozyme banding patterns or zymogram patterns were read by a spectrophotometer equipped with a gel scanner. The results indicated that zymogram patterns of both isozymes in tested plants were specific to their genetic background, and the banding patterns among cultivars, varieties and parts of plants were found to be different. The disc PAGE of plant isozyme is proved to be a valuable tool for discrimination of plant varieties and plant genetic variations.
