Murine model of acute toxoplasmosis for the evaluation of drug efficacy

The mouse is the animal of choice for the in vivo evaluation of the efficacy of antitoxoplasma chemotherapy as infection with even a single parasite of the virulent RH strain is unavoidably lethal. In the model we developed Swiss Webster mice were intraperitoneally (i.p.) inoculated with suspensions containing 10**2 - 10**6 semipurified RH Toxoplasma gondii tachyzoites. Drug efficacy was assessed on the basis of mouse survival rates and mean survival times. The model was tested with clindamycin (400 mg/kg/day), sulfamethoxasole (375 mg/kg/day) and co-trimoxasole (480 mg/kg/day). Differences obtained in the protective effects of these drugs as well as in the results of additional procedures performed to determine the outcome of infection showed that the proposed murine model is a sensitive and versatile model system for the evaluation of drug efficacy in acute toxoplasmosis.
