Influence of microinjection on developmental potential of rabbit embryos in vitro

Object of the study was to investigate the effect of microinjection of foreign gene on developmental potential (cleavage rate) of cultured rabbit zygotes and rate of foreign DNA integration (PCR). Totally 970 zygotes of pronuclear stage were collected from 75 donors. The WAP-hPC gene was injected to pronucleus of 515 eggs, and 425 eggs were used as intact control. After microinjection, the zygotes were cultured during 72 h. Microinjection significantly decreased cleavage of zygotes (21.5 vs. 58), as well as embryonic development up to morula/blastocyst stage and increased the proportion of degenerated eggs. Summarized PCR data indicates that 6.6 (34 of 515) of all the microinjected zygotes, having reached the morula or blastocyst stage, demonstrated the positive signals for hPC gene integration.
