Identification of legumin-like protein cDNA clone from cDNA library of developing buckwheat seed

cDNA library of the buckwheat (Fagopyrum esculentum Moench) seed in mid-maturation stage of development has been constructed by RT PCR approach. Clones obtained were divided into two groups - 80% of them were seed-specific (group 1), while 20% were non-specific for the seed (group 2). Among group 1, clone pBF 349 showed high homology with legumin-like storage protein cDNA clones from a variety of plant species. Expression pattern of mRNA in different buckwheat tissues, using pBF 349 as a probe in Northern analysis, confirmed its seed specificity.
