Structure and editing of a gene coding for ribosomal protein S2 in wheat mitochondria

We have identified a wheat mitochondrial gene that codes for a protein showing homology to bacterial ribosomal protein S2 (RPS2). This gene is located upstream of the wheat nad9 gene, and is transcribed from the opposite strand as a single mRNA of 1.7 kb. The RNA extremities were mapped by primer extension and RNase protection. The 5'-end of the transcript maps at the level of a sequence that matches the consensus for plant motochondrial promoters, implying that the 1.7 kb mRNA corresponds to a primary transcript. The RNA sequence was determined from cDNA clone obtained by RT-PCR. Several differences to the genomic sequnce were found, resulting from C to U RNA editing. These differences improve the amino acid homologies of the deduced protein sequence with other RPS2 protein sequences. Therefore, editing must have been essential for the expression of a functional protein.
