Effect of gamma-rays irradiation in seed of mungbean (Vigna radiata (L.) Wilczek) composition of media on shoot regeneration of explants from node of cotyledon

英语. Study the effects of gamma-rays irradiation and composition of media on shoot regeneration of explants from node of cotyledon of mungbean Walet variety have been conducted. The explants derived of irradiated seeds of mungbean Walet variety have been conducted. The explants derived of irradiated seeds of 10-20 Gy of gamma rays were planted in the 0.7 percent agar solution. One day after planting in the agar media the embryo axis of germinate seeds were removed and the node of cotyledon were cultured in the regeneration media as explants. The results showed that shoot regeneration was influenced by media composition and the doses of gamma rays irradiation in seed. In the MURASHIGE and SKOOG medium which contain of BAP or 2-iP or Kinetin with 3 ppm concentrate respectively the explants could produced 100 percent of shoots. However, the highest number of produced shoot (3 shoots) was showed in the medium which contained of BAP. The medium with 1 ppm concentrate of BAP could produced 100 percent shoot regeneration and the maximum number of shoots (4 shoots) per explants was shown in with 5 ppm concentrate of BAP. The effectivity of BAP for shoot regeneration could be improved by enrichment of 12 ppm Ag2SO4 in the media. 显示更多 [+] 显示较少 [-]印度尼西亚. Sudah dilakukan penelitian mengenai pengaruh iradiasi sinar gamma pada benih kacang hijau varietas Walet dan komposisi media terhadap regenerasi pucuk ruas kotiledon. Eksplan berasal dari benih kacang hijau yang diiradiasi sinar gamma dengan dosis 10-30 Gy ditanam pada larutan agar 0,7 persen. Sehari sesudah tanam kedua kotiledon diambil dan embrio aksisnya dibuang. Eksplan tersebut kemudian dikulturkan pada media regenerasi dengan berbagai komposisi. Hasil penelitian menunjukkan bahwa banyaknya pucuk yang teregenerasi dipengaruhi oleh komposisi media dan dosis iradiasi sinar gamma pada benih kacang hijau. Media MURASHIGE dan SKOOG (MS) yang mengandung BAP atau 2-iP atau kinetin masing-masing dengan konsentrasi 3 ppm dapat menghasilkan pucuk 100 persen dari eksplan yang digunakan, akan tetapi jumlah tertinggi (3 pucuk) dihasilkan dengan penambahan BAP. BAP pada konsentrasi 1 ppm sudah dapat menyebabkan terjadinya regenerasi pucuk buku kotiledon 100 pesen, sedangkan jumlah pucuk maksimal (4 pucuk) dicapai dengan konsentrasi 5 ppm. Efektifitas BAP pada regenerasi pucuk buku kotiledon dapat dinaikkan dengan penambahan Ag2SO4 sebanyak 12 ppm. Iradiasi sinar gamma pada dosis 10 dan 20 Gy pada benih kacang hijau dapat meningkatkan regenerasi pucuk buku kotiledon kacang hijau varietas Walet
