Raising of new cultivar "Saga-Otome" by crossing of amphidiploidy Allium wakegi x A. fistulosum

Chromosomes of Allium wakegi Araki, Allodiploidy plant (2n=16), were doubled by callus culture, and amphidiploidy plants (2n=32) were crossed with A. fistulosum L. (2n=l6) to raise a new cultivar. Shoot tips of "kumamoto-zairai" were cultured on MS medium supplemented with 1 mg/l 2,4-D. After 2 months, embryogenic calli were induced from basal plate, and removed to new medium supplemented with 1 mg/l kinetin. 42 number of plantlets were regenerated through embryogenesis. In these plantlets, 9s were tetraploidy variants, and 1 was aneuploidy variant (2n=31). Amphidiploidy plant was restored female fertility, but not restored male fertility. "Omura-natunegi" was pollinated to amphidiploidy plant, and superior plant was selected in these hybrid seedlings. After characteristic examination, it was established to new cultivar, named "Saga-otome" is triploidy plant (2n=24) and propagated by bulblets. It shows light dormancy and heat tolerance, so suits to summer cultivation
