Presence of melatonin receptor in the anterior lobe of pituitary of the hen

By the use of radioligand binding assays, the membrane fraction of anterior lobe (pars distalis) of pituitary of the hen was found to contain melatonin binding component having characteristics of a receptor such as high affinity, limited capacity, reversible binding, and binding specificity. The equilibrium dissociation constant (Kd) obtained by Scatchard analysis and by kinetic analysis, expressed as moles per litter, was of the order of 10(-9) and the maximum binding capacity (Bmax) was of the order of fmol per mg protein. The value of Kd was not different between laying and non-laying hens, while the Bmax value was greater in laying hens. The results suggest that a melatonin receptor is present in the anterior lobe of pituitary of the hen and that the receptor binding may change in relation to egg-laying
