Screening mulberry (Morus spp.) genotypes for adventitious bud formation and plant regeneration from immature leaf culture

To select mulberry genotypes having a high capability of forming adventitious buds and regenerating, 287 genotypes (Morus bombycis, 89; M. alba, 57; M. latifolia, 104; other species, 37) were screened by culturing immature leaves in MS medium containing 4.4 mu-M benzyladenine for adventitious bud induction and in MS medium containing 1.1 mu-M indoleacetic acid for root induction. Adventitious buds formed in 121 genotypes (M. bombycis, 28; M. alba, 27; M. latifolia, 47; other species, 19). A total of 83 genotypes developed shoots from adventitious buds, and 55 (19%) genotypes rooted and regenerated whole plants (M. bombycis, 14; M. alba, 15; M. latifolia, 19; other species, 7). Adventitious bud formation and plant regeneration from immature leaf cultures varied considerably by genotype. Several genotypes having a high capability for adventitious bud formation were selected: for example, Kanadasansou A showed the highest percentage (72%) of adventitious bud formation, while Oyutaka formed the largest number (6.3) of adventitious buds per explant and showed a high percentage of shoot formation. Adventitious buds were repeatedly formed by refreshing the culture in which the regenerated shoots were removed and the original immature leaf explants were recultured, indicating that the adventitious bud induction system is suitable for the in vitro propagation of the mulberry
