Development of tissue culture technique for mangosteen rapid propagation

This study was conducted at the Tissue Culture Laboratory, Crop Science Division, USMARC, USM, Kabacan, Cotabato [Philippines] from November 1999 to March 2000 to evaluate the effect of Benzyl Adenine on the growth and plant regeneration of mangosteen, find out the most appropriate culture medium for early proliferation and plant regeneration, determine the effect of light and dark condition in the growth and early proliferation and plant regeneration, and to evaluate the possible interaction with culture media supplemented with Benzyl Adenine at various levels. The experiment was carried out in a Completely Randomized Design (CRD) in factorial with eight treatments replicated ten times. Benzyl Adenine at various levels significantly affects the growth and plant regeneration of mangosteen. Increased in growth enhanced regeneration was noticeable in culture as indicated in terms of the final number of shoots 40 DAI, shoot increment, degree of plant proliferation and stem girth compared to the control. However, Benzyl Adenine showed that significant effect on the number of days to initial shoot formation, number of shoots to initial shoot formation, number of leaves, length and width of leaves. The length and width of leaves was affected by the light condition significantly. Culture media and light condition interaction effects was observed also in the length of width of leaves. The most appropriate culture medium of early proliferation and plant regeneration of mangosteen is exhibited by MS [Murashige Skoog] Media with the addition of 5 ppm Benzyl Adenine
