Physico-chemical properties and brine shrimp cytotoxicity of bovine and porcine sera [gamma]-irradiated at virus-inactivating doses

Transmission of viruses and other pathogens by animal sera can potentiate considerable risks for humans and animals particularly when the serum is being used for the production of pharmaceutical products, such as vaccines and in other related R and D activities in cell biology. For this purpose, the possibility of exposing sera derived from local abattoirs to high doses of gamma radiation for both sterilization and virus-inactivation is currently being study. The radiolytic response of bovine and porcine sera exposed to 3, 6, 9, and 12 kGy gamma radiation was investigated based on the physico-chemical parameters of viscosity, total protein content and pH. Significant decline in pH, viscosity and total protein have been observed relating to the radiolytic phenomena of main-chain scission, protein degradation and the release of amino acid groups. Despite the grossly altered physico-chemical properties, cytotoxic radiolytic products from the sera were not detected based on the microplate brine shrimp (Artemia salina L.) assay. An important findings warrant the biosafety of radiation-sterilized animal sera considering that viral pathogens, i.e. FMDV and other zoonoses, are readily inactivated at doses of 9-12 kGy. Irradiation can therefore be effectively utilized to sterilize sera that will later be developed as a safe, cheaper and more available substitute to commercial serum products. Further work on clonogenicity to a battery of human and animal cell lines is still in progress
