Production of pullulanase and beta amylase by Bacillus subtilis

A strain of Bacillus sp. was isolated from soil and identified as Bacillus subtilis. Specific carbon an nitrogen sources were required for growth and yield of enzymes. The maximum production o pullulanase and Beta-amylase was found with ammonium oxalate and ammonium di- hydrogen phosphate, respectively. Maximal enzymes activities were achieved after 48 hours of shaking cultivation at 37 Centigrade and at initial pH 6.0. Pullulanase and Beta-amylase synthesis was significantly enhanced by liquefied potato and corn starches as well as dextrin, fructose, ma/t extract, casein hydrolyzate.
