Production of astaxanthin from Haematococcus sp.

New strain of Haematococcus sp. isolated from Huay Kha Khaeng Wildlife Sanctuary, Uthaithani Province was identified as H. lacustris subsp. siamensis. It could grow well under autotrophic condition at 30 deg C in MCM medium and was able to produce 30 mg/l of astaxanthin which was comparatively higher than other 7 Haematococcus strains obtained from culture collections. In the study of growth and astaxanthin production, MCM medium was modified by increasing concentrations of some inorganic nutrients and vitamins. When ferric chloride concentration was raised from 0.0244 mg/l to 12.49 mg/l and small amounts of biotin was added the astaxanthin accumulated increased from 30 mg/l to 55 mg/l, a gain of 25 mg/l as compared to those cultivated in original MCM medium. In contrast addition of vitamin B12 to the medium did not enhance both growth and astaxanthin production. Under mixotrophic growth condition at 20 deg C in basal medium containing acetate as carbon source, the adjustment of nutrient composition did not improve growth and astaxanthin production, however increase of light intensity shown little positive effect. Elevation of cultivation temperature from 20 deg C to 30 deg C could also enhance astaxanthin production in one Haematococcus strain. When the algae was cultivated in 1 L agitated bioreactor containing basal medium, growth and astaxanthin production was reduced. Outdoor cultivation of H. lacustris subsp. siamensis in 5-liters rocking trays under natural condition with 20 percent inoculum size using modified MCM medium resulted in production of astaxanthin at the rate comparable to cultivation in laboratory, however scale up to 100 L raceway cultivation was not successful due to contamination of other algae.
