While the reason of variation in rooting of semi-hardwood olive cuttings of hard-to-root (Domat) and easy-to-root (Gemlik) cultivars and rooting mechanism have been investigated, total sugar and starch level were analyzed. Total sugar content was generally higher in Domat. The cuttings of 1996 (on year) and 90. days have less total sugar than 1995 (off year) and 15. days. At the same time, the endogenous starch content of cuttings was found to be lower in 1996, while the metabolic activity in root and stem formations was higher during this year. Generally, Domat and 15th day cuttings have higher levels of starch, however there are no significant differences between time periods in Domat (in contrast to Gemlik), instead of root formation, dense callus formation is observed
