Construction of a series of pGEM/cryIA mutants uni-directionally deleted at the 5' end adjacent to the insert

英语. CryIA gene encodes toxin with highly specific insecticidal activity to larvae of lepidoptera such as pod borer of cocoa and leaf-chewing caterpillar of oil palm. The 2-kb cryIA fragments encoding toxin domain cloned in cloning vector pGEM-T will be sequenced. There are two strategies that may be applied to sequence the entire coding region of the gene, sequencing by using primer walking which requires at least four DNA primers, or using a series of corresponding deleted mutants that needs only one primer. In case of the availability of the primer depends on imports, the second strategy is considered more efficient than the first one since the primer for the following sequencing in the first strategy has to be designed based on the result of previous sequencing. This research was aimed to construct a series of pGEM/cryIA mutants uni-directionally deleted from the 5' end at some extents in conjunction with sequencing of the cryIA insert. The construction was conducted with Erase-A-Base system from Promega. To generate linearized plasmid digestible only from the 5' end adjacent to the cryIA insert, the recombinant plasmid was double digested with ApaI and NcoI. The result of enzymatic deletion indicated that with enzyme/DNA ratio 40 percent higher than the standard procedure, the rate of exonuclease digestion was correspondingly higher. Electrophoretic examination of the plasmids isolated from selected bacterial clones demonstrated that the construction of the recombinant mutants was expectedly completed. Clones generated from longer exonuclease digestions were proven to carry smaller mutated plasmids

Fragmen gen cryIA penyandi domain toksin yang berukuran 2 kb dan telah diklon di vektor kloning pGEM-T akan dianalisis sekuennya. Ada dua strategi yang dapat diterapkan dalam penentuan sekuen daerah penyandi pada gen tersebut, yaitu sekuensing menggunakan teknik primer berjalan yang memerlukan paling tidak empat DNA primer, dan menggunakan beberapa mutan terdelesi yang membutuhkan hanya satu primer. Dalam keadaan ketersediaan primer tergantung pada impor, pendekatan kedua dianggap lebih efisien daripada yang pertama karena primer untuk sekuensing berikutnya pada strategi pertama harus dirancang berdasarkan hasil sekuensing sebelumnya. Dalam rangka penentuan sekuen gen cryIA, penelitian ini bertujuan untuk membuat serangkaian mutan pGEM/cryIA yang terdelesi searah dari ujung 5' dengan berbagai tingkat. Percobaan dilaksanakan dengan menggunakan kit Erase-A-Base System dari Promega. Untuk menghasilkan DNA linier yang dapat didelesi hanya dari ujung 5'-nya, plasmid rekombinan tersebut didigesti ganda dengan ApaI dan NcoI. Hasil delesi enzimatis menunjukkan bahwa dengan nisbah enzim/DNA sebesar 40 persen lebih tinggi daripada prosedur standar, laju digesti oleh eksonuklease juga lebih tinggi secara proporsional. Pengujian elektroforesis plasmid yang diisolasi dari klon bakteri terseleksi menunjukkan bahwa perakitan mutan rekombinan tersebut berhasil sebagaimana diharapkan. Klon yang berasal dari digesti eksonuclease yang lebih lama terbukti membawa plasmid termutasi yang ukurannya lebih kecil
