The effect of elicitor derived from Pythium aphanidermatum [Edson] Fitzp. on Ajmalicine content of Catharanthus roseus [L.] G. Don callus cultures

Callus of Catharanthus roseus (L.) G. Don was induced from leaf segment, and grew optimally on Zenk medium with the addition of 2.5 micrometer NAA and 10 micrometer BAP. Callus from the third subculture was elicitated with elicitor derived from autoclaved cell of Pythium aphanidermatum. The concentrations of elicitor tested were 0.00, 0.05, 0.05, 1.00 and 5.00 mg dry weight/ml, and the harvesting times were 0, 18, 36, and 72 hours after elicitation. Quantitative and quantitative analysis were performed on callus, leaves, and roots of C. roseus. Our study indicated that ajmalicine content was influenced by concentration of elicitor and harvesting time. A significant increase of ajmalicine content (440.572 approx. 9.687 microgram/g dry weight) was achieved by the addition of elicitor of 0.50 mg dry weight/ml after 36 hours, whilst the control produced only 245.936 approx. 4.351 microgram/g dry weight. There was no ajmalicine detected from leaves, whilst root extract contained only 86.0 microgram/g dry weight. This study showed that there was a significant increase of ajmalicine content in C. roseus callus cultures after being challenged with P. aphanidermatum elicitor
