Comparison of the bobcock and detergent methods of butterfat analysis

The detergent, Triton X-100 (octyl phenoxy polyethoxy ethanol) gave consistently lower milk fat tests of 0.01, 0.04 and 0.14 percent than the Babcock method for milk from Holstein x Red Sindhi, Brown Swiss, and Murrah buffalo cows, respectively. The corresponding tests of the same samples with Triton N-101 (nonyl phenoxy polyethoxy ethanol) were 0.10, 0.13 and 0.35 percent lower than the Babcock tests. However, the differences are not significant. These detergents are not suggested for testing skim milk of 0.50 percent or less fat content. For testing fat in ice cream, they are as good as the Babcock method. Although not significant, milk fat tests of the same samples done by two analysts differed by as much as 0.22 percent for the high fat milk. The daily milk fat tests in seven consecutive days of milk from the same cows varied significantly at one percent probability level. Milk production also varied significantly. Savings in sample and reagent are effected with the use of 10 ml high-fat carabao or Murrah buffalo's milk and 10 ml sulfuric acid is a standard 8 percent Babcock test bottle. Besides, the possibility of errors in dilution of high-fat milk is removed. By proportion, milk fat tests should be expressed in terms of 17.6 ml milk.
