Antimicrobial metabolites from microorganisms associated with sponges and tunicates

The alarming rise of infectious diseases caused by bacteria rapidly developing resistance towards traditional antibiotics had led to the search for sources of new antimicrobial agents. One relatively untapped resource for such agents is marine orgnanisms, less than 5% of which have been cultured and identified. This research aims to isolate novel antibiotic from marine microorganisms. Microbial epiphytes and symbionts of sponges and tunicates collected by SCUBA from Davao and Zamboanga, Philippines were isolated. The microbial isolates totaled 385:167 from 78 Davao sponges and tunicates and 218 from 100 Zamboanga sponges and turnicates. Culture broths of 102 isolates were tested for antimicrobial activity using the Kirby-Bauer disc diffusion assay and 35 were further tested using the Microplate Alamar Blue Assay (MABA). Of the 80 isolates tested, 2 were found active against Vibrio harveyii and out of 32, 4 have activity against Escherichia coli 11775 and only one has activity against Klebsiella pneumonia subsp. pneumoniae 13883. Eleven were active against Aeromonas hydrophila 7966, 20 against Pseudomonas aeruginosa 10145 and 43 against Staphylococcus aureus subsp. aureus 12600. Five isolates were also found active against Mycobacterium tuberculosis at 50? g/ml of the crude capsular extract through the Microplate Alamar Blue Assay. Bioassay-guided purification and the optimization of antibiotic production of the four promising isolates, Z143-1, Z39-1, Z105-2 and 68-1 are underway. The culture broth of Z143-1 was initially extracted by Kupchan partitioning and the hexane fraction tested positive against S. aureus in the TLC overlay assay
