Isolation and characterization of fruit amylase from melon

Ripe and fungus free melon (Cucumis-melo) was used for extraction, purification and characterization of amylase. Enzyme activity was determined only in ripe specimens. Two types of amylases, amylase-I and amylase-II were isolated from the crude extract of ripe specimen on sephadex G-75 chromatography. The enzyme was purified by gel filtration. Two protein peaks were obtained. The first protein peak did not contain the enzyme. The enzyme was present under second protein peak. Purified enzyme had maximum activity at 50 degree centigrade and 8.5 pH. The Km value of the enzyme was 4.58 g/ml.
