The low yields of tef are mostly attributed to its susceptibility to lodging. The primary goals of this project are to 1) construct a comparative genetic map for tef and determine genetic relationships with other grasses and 2) identify loci or candidate genes attributing lodging resistance. There are no existing maps of tef that can be compared with other cereals. We are using a recombinant inbred (RI) population derived from an interspecific cross, E. tef cv. Kaye Murri X E. pilosa cv. 30-5, developed at the Debre Zeit Agricultural Research Center in Ethiopia, for the purpose of comparative genetic map construction and for the analysis of quantitative trait loci (QTL) affecting lodging. Currently we are using the grass anchor probes, which originated from rice, maize, barley, oats and millets. In addition, we are using microsatellities from wheat and maize as well as EST derived SSRs from several grass species. Inter-Simple Sequence Repeat amplification (ISSR) technique is being used to saturate the map for use in identifying QTL associated with lodging and other important traits. We identified a number of candidate genes that may play a role in either dwarfing or lodging resistance and these will be mapped in tef to test their ability to improve lodging related traits in tef.
