The influence of senna leaves water extract on rat liver enzymes activity

Senna (Cassia senna L., Cassia acutifolia) is a well-known drug used in the treatment of constipation. Its purgative action is due to the presence of sennosides. As there were some suggestions about possible carcinogenic or toxic effect on liver and colorectal cells in chronic use of sennosides, the aim of our assay was to examine the influence of senna water extract on rat liver enzymes activity and systems. Wister rats were used and divided into 4 groups according to the duration of per oral senna extract intake (control (no senna intake) and 3 groups which were receiving 1 mL/day during 1, 3 and 7 days). Senna leaves water extract (maceration), was consisted of about 0.32-0.35 mg of sennosides B/mL. Results showed significant reduction of weight gain of the animals after 1st and 3rd day, while it was less on 7th day. The activity of glutathione-peroxidase, as well as lipid peroxidation was significantly increased, espacially on 7th day, while the content of glutathione was decreased earlier, on 3rd day. The activity of liver peroxidase was increased after the first application of senna extract and reached the maximum value on 7th day. During the whole examined period, the activity of liver catalase did not significantly change. These results may suggest on possible interactions and potentiation of oxidative action of other drugs/substances taken concomitantly during senna intake.
