Detection of chloramphenicol residues in animal products by ELISA (Enzyme Linked Immunosorbent Assay)

英语. The occurrence of chloramphenicol (CAP) residues in animal products may cause serious problems to the consumers, such as aplastic anemia. A sensitive and specific method is urgently needed to detect residues in animal products. A direct ELISA method was developed using chloramphenicol - bovine serum albumine (CAP-BSA) as antigenic compound, and antiserum CAP was labelled with horseradish peroxidase (HRP) as antibody. CAP-BSA was synthesized from CAP sodium succinate and BSA by mixed anhydride, while the antiserum CAP was labelled with HRP by periodate conjugation technique. A dilution of 1:200 of CAP-BASE and 1:125 dilution of antiserum CAP labelled with HRP gave limit detection of 0.01 ng/ml, while recovery study of CAP from animal products ranged from 91.70 to 98.17 percent

Adanya residu kloramfenikol (CAP) dalam produk peternakan dapat menimbulkan efek samping yang cukup serius kepada konsumen seperti aplastik anemia, sehingga diperlukan suatu metode analisis yang cukup sensitif dan spesifik untuk mendeteksi residu CAP. Telah dilakukan pengembangan metode ELISA langsung menggunakan CAP-bovine serum albumin (CAP-BSA) sebagai antigen dan antiserum CAP yang dilabel dengan enzim horseradish peroxidase (HRP) sebagai antibodi. CAP-BSA disintesis dari CAP natrium suksinat dan BSA mempergunakan reaksi anhidrida campuran, sedangkan antiserum CAP diberi label enzim HRP dengan teknik konjugasi periodat. CAP-BSA dengan pengenceran 1:200 dan antisera CAP berlabel HRP dengan pengenceran 1:125 menunjukkan limit deteksi 0,01 ug/ml, sedangkan uji perolehan kembali CAP dari produk ternak berkisar antara 91,70-98,17 persen
