Optimization studies on glucoamylase produced from banana stalks

The project was planned for the maximum yield of glucoamylase from waste banana stalks by Bacillus subtilis. Different parameters like fermentation period substrate water ratio, effect of pH, effect of temperature, incubation period and varying concentration of peptones alongwith effect of surfactants like sodium dodecyl sulphate (SDS) and Tween-80 were aloe studied. The growth media of banana stalks powder was fermented by Bacillus subtilis through solid substrate fermentation (SSF). The pH was maintained at 7 and placed at 35 degree C for 24 hours of incubation period. After 24 hours, 100 ml of sodium acetate buffer of pH 4.8 was added to the flasks and then shacked at 150 rpm on an orbital shaker for 30 minutes. It was then filtered and the filtrate was centrifuged 10000 rpm at -10 degree C for 15 minutes. The supernatant was then subjected to enzyme assay. The maximum activity of glucoamylase (8.425 U/ml/min) recorded after 24 hours of solid state fermentation of the growth medium containing 5g of banana stalks powder having 70% moisture, 0.2% peptone, 0.1% SDS and 0.1% Tween-80. It was observed that the addition of s urfactants like SDS and Tween-80 enhanced the production of glucoamylase. Peptone have an enhanced effect on enzyme production because in proteins several N- containing amino acids are present which are a good source of nitrogen and have enhance effect. Glucoamylase production by Bacillus subtilis from banana stalks powder medium gave reasonable yields of the enzyme and ma be economical.
