[Obtention of embryos through anther culture in Brassica oleracea [Portugal]]

Six populations of Brassica oleracea var. costata D. C. (Vila Verde, Sampaio I, Sampaio II, Santo Estevao, Passos e Vidago) were studied to obtain embryos by anther culture. Four media (B5 + 0.1 mg/l NAA + 0.1 mg/l 2.4 - D; B5 + 0.1 mg/l NAA + 0.5 mg/l 2.4 - D; B5 + 0.5 mg/l NAA + 0.1 mg/l 2.4 - D; B5 + 0.5 mg/l NAA + 0.5 mg/l 2.4 - D) with different concentrations of two auxins (2.4-D and NAA) were used. The light effect in the embryos induction was also analysed. From the results obtained it can be concluded that the auxin 2.4-D gave better results in the production of embryos, increasing the number with the higher auxin concentration. The effect of the genotype is statistically significant (P0.001). The Santo Estevao population performed best with 97.14% of the total of embryos obtained and with an average of 161.9 embryos / 100 anthers. The number of embryos registered from the anthers developed in the dark until the first appearence of embryos, was lower than from the anthers developed with light.
