Laboratory-scale production of antifungal substances from Bacillus subtilis

Laboratory-scale production of antifungal substances from Bacillus subtilis TISTR 1 was studied in 5-litre fermenter, using 10 percent inoculum. The optimum conditions in the fermenter were: agitation rate 300 rpm, aeration rate 1 vvm, controlled temperature at 35 deg C, and 48 hr culturing period. Comparing two culture media, trypticase soy broth supplemented medium could provide better growth of bacteria cells than did waste soybean extract medium. The culture broth was subsequently either filtered through sterilized membrane or heated at 120 deg C for 5 minutes. When tested against 11 strains of plant pathogenic fungi, both types of the culture broth completely inhibited 6 strains of those fungi.
