Cryopreservation of banana germplasms for collection

Success of cryopreservation or preservation of living tissues at temperature below -130 deg C depends on genotype of the organism. A Thai banana, TMP 04, obtained from the wild, was grown in vitro in modified MS medium at the BAP concentration of 10*[-4) M to induce cauliflower-like meristems. High sucrose+glycerol followed by pretreatment of cryoprotectant mix of glycerol + ethylene glycol + DMSO were necessary steps before liquid notrogen (-196 deg C) exposure. After cryotreatment for at least 30 min., samples were recovered in liquid medium + sucrose, and were later transferred to grow in the dark for about one month. TMP 04 was able to survive the cryotreatment and continued to grow upon changing to routine culturing at 25 deg C with lighting.
