Preliminary study on protoplast culture of mungbean (Vigna radiata L.)

Protoplasts were isolated from the hypocotyls of mungbean (Vigna radiata L.) seedlings by incubating in the enzyme solution containing 0.5 percent cellulase R-10, 1.0 percent driselase, 0.05 percent pectolyase Y-23, 0.5 M sorbitol and 5 mM CaCl2 for 16 h. The isolated protoplasts were cultured in the MS liquid medium supplemented with 1.0 mg/lt BA, 1.0 mg/lt NAA, 0.5 mg/lt 2, 4-D and 14 percent sucrose, After one week of culture, the protoplast derived cells were embedded in 0.6 percent agarose, The first cell division was obtained in 4-7 days of culture and microcolonies were formed in one month. The protoplasts from dark-grown seedling seemed to survive better and grow faster than those from light-grown seedling.
