Purification and characterization of N-acetylgalactosamine specific lectin from Champaada (Artocarpus integer) seed

N-acetylgalactosamine specific lectin from Champaada (Artocarpus integer) seeds has been purified by ammonium sulfate precipitation and affinity chromatography. It is assumed to be a trimer having the molecular weight of 48,000 with subunits molecular weight of 14,800 and 17,300 as determined by gel filtration and by gel electrophoresis in the presence of sodium dodecyl sulfate. Amino acid analysis have shown that purified lectin is low in arrginie, histidine and methionine and absent in cystine residue. Champaada lectin agglutinated erythrocytes of various animals except of goat and chicken. The hemagglutinating acitivity was not effected by divalent cations and SH reagent. The lectin is stable at neutral pH and thermally stable.
