Identification of DNA finger printing of Indonesian taro (Colocasia esculenta L. Schott) with RAPD DNA technique: primary screening and optimalization of PCR edition

DNA finger printing technique has been widely used in the analysis of genetic variability of both plants and animals. One of the techniques that can be used to obtain DNA finger print is RAPD (Random Amplified Polymorphic DNA) technique, a molecular technique based on PCR technology. The quality of amplification products depend on several factors, such as MgCl2, and DNA concentrations, primer and PCR condition. In this study, an experiment was conducted to find out appropriate PCR condition to amplify taro DNA using random primers. Three PCR conditions have been tested and one of them successfully amplified taro DNA with good results. Of the 12 primers tested, 9 of them (OPB-01, OPB-04, OPB-05, OPB-06, OPB-07, OPB-10, OPB-15, OPB-16, and OPB-20) were able to produce amplification products. Primers OPB-01, OPB-04, OPB-05, OPB-06, OPB-07, OPB-15, and OPB-20 produced polymorphism bands on some of the samples studied. The results therefore indicated that the technique can be used to evaluate genetic variability of taro at molecular levels and identify additional markers in selection and breeding of the crop
