Antagonism of detomidine induced sedation, bradycardia and ruminal tympany in atropinised buffalo calves

Male buffalo calves (36-4 groups of 9 calves each) were injected IM and IV detomidine @ 50 æg/kg in group 1 and 2, 3 times dose @ 150æg/kg in group 3 and 5-times dose @ 250 æg/kg in group 4. At the maximum depth of sedation they were given IV normal saline 2 ml (control), yohimbine @ 0.125 mg/kg or atipamezole @ 50æg/kg IV in each group of animals. In calves given standard dose of detomidine IM, the MAT recorded was 60.20ñ2.41, 2.26ñ0.27 and 0.90ñ0.07 min in control, yohimbine and atipamezole administrated animals the MST was 78.72ñ4.23,3.68ñ0.23 and1.68ñ0.13 min while MWT was 108.00ñ5.22,4.02ñ0.59 and 1.92ñ0.19 min. In IV groups MAT was 54.82ñ3.28, 2.28ñ0.39, 0.09ñ0.13, MST was 65.00ñ5.18, 4.94ñ0.53 and 1.36ñ0.12 min and MWT was 100.00ñ3.72, 5.46ñ0.96 and 1.48ñ0.14 min in control yohminbine and atipamezole, respectively. The overdoses of detomindine (3X and 5X) were also effectively antagonized by yohimbine and atipamezole. Detomidine induced hypothermia, bradycardia, respiratory depression and ruminal atony were completely antagonized by yohimbine and atipamezole. Atipamezole caused quicker and more effective reversal than yohimbine. Thus atipamezole or yohimbine can be safely used as useful antidote for detomidine overdose in buffalo-calves.
