Turbidometric evaluation of antibacterial activity of lower alcohols against Escherichia coli

Antibacterial activities of lower alcohols against Escherichia coli IFO3972 were evaluated by measuring the change in the turbidity of the growth medium with the bacterial multiplication (turbidometry). The turbidity curves shifted toward a longer incubation period and the slope became gentle as the alcohol concentration increased. The growth rate and the minimum inhibitory concentration (MIC) were estimated by the application of the inhibition kinetic model proposed by Takahashi et al. for calorimetric evaluation. Drug potency (antibacterial efficacy) curves of lower alcohols could be drawn based on the Kinetic parameters, and the growth rate and MIC could be determined. The obtained antibacterial activity of lower alcohols increased with increases in the 1-octanol/water partition coefficient (P) values and water-accessible surface areas (ASA). Especially, the order of methyl groups arranged according to their spherical shape, MeOH-EtOH-i-PrOH-t-BuOH, had a higher linearity against log(P) and ASA than the order according to the elongation of the normal carbon chain.
