The role of calcium-dependent protein kinase (CDPK) of potato microsomal fraction in signal transduction of the hyphal cell wall component and the suppressor from Phytophthora infestans (Mont.) de Bary

We investigated the localization of calcium-dependent protein kinase (CDPK) in potato microsomal fraction (MF), and the effect of the hyphal cell wall component (HWC) and the suppressor glucan (suppressor) of Phytophthora infestans (Mont.) de Bary on hypersensitive resistance response (HR). We also studied the activity of CDPK from MF in the absence and the presence of staurosporine, a kinase inhibitor and okadaic acid, a phosphatase inhibitor. A 70 kDa CDPK was detected in potato MF by using western immunoblot analysis. HWC and suppressor stimulated the kinase activity of the MF within 5-10 min after the application. In the presence of staurosporine, the HWC stimulated the kinase activity of the MF but not the suppressor. In the presence of okadaic acid the HWC inhibited and the suppressor stimulated the kinase activity of the MF. These results suggest that the HWC and the suppressor may interact with the potato kinase(s) in the MF resulting in the signal of HWC for the occurrence of HR and in case of suppressor for the inhibition of that. We suggest that the signal cascade of the HWC and the suppressor of P. infestans may be different in incompatible and compatible interaction in potato and P. infestans interaction, respectively.
