Effect of nurse culture and conditioned medium on colony formation from Oenanthe stolonifera protoplast

In order to apply the culture of asymmetric somatic hybrid cells between Oenanthe stolonifera 'Shimane-midori' and other Umbelliferae plants, the effect of nurse culture and conditioned medium on colony formation from O. stolonifera protoplasts was studied. When O. stolonifera protoplasts were cultured with celery 'soup-celery' calli as nurse cells, colony formation progressed in the early stage of culture, and no impediment occurred thereafter. Conditioned medium, prepared from celery suspension culture, also increased the colony formation in the early days of O. stolonifera protoplast cultures. When protoplast cultures using conditioned medium were continued to 60-90 days, colony formation was impeded. However, when conditioned medium was exchanged for fresh protoplast culture medium at 10-30 days after culture, colony formation was not impeded until 90 days after culture. Between days 7 and 28, the longer the period of celery suspension culture for preparing conditioned medium, the greater the number of O. stolonifera protoplast colonies that formed. When the conditioned medium was stored at -20degC to room temperature or boiled at 121degC, the conditioning effect on colony formation was maintained.
