Micro-propagation of sugarcane through bud culture

Improvement in wild varieties of sugarcane (Saccharum officinarum L.)through advanced techniques of tissue culture has not so far been reportedfrom NWFP. The present study is an attempt to evaluate two wild relativesErianthus 3854 and SES 089 for medium standardization, sterilization andregeneration to explore the potential of these wild lines in further breedingprogram. Buds of these lines were sterilized with different chemicals anddurations. The best results (60 and 75 %) were obtained when buds of bothsugarcane varieties were treated with 0.1% HgCI (Mercuric Chloride) for 10minutes. Maximum shoot development (83.3 % with average shoot length 3.7 cm)was achieved on MS (Murashige Skoog) medium supplemented with 5.0 mg/IBAP (Benzyl Amino Purine) and 1.0 mg/l GA3 (Gibberillic acid), while theminimum shoot development (29.1 % with average shoot length 6.7 cm) wasrecorded for Erianthus 3854 on medium containing 5.0 mg/I BAP and 0.25 mg/lGA3. For SES 089 maximum shoot development (62.5 % with average shootlength 3.2 cm) was obtained on MS medium containing 0.5 mg/l BAP and 1.5mg/l GA3, while the minimum shoot development (20.8 % with average shootlength 1.9 cm) was recorded for the combination of 0.5 mg/l BAP and 0.25 mg/lGA3.
