Effect of growth regulators on embryogenic callus induction in narcissus (Narcissus L.) Carlton culture

Effect of auxin (Picloram or 2.4-D) and cytokinin on embryogenic callus induction and somatic embryos formation in narcissus shoot ovary explant culture was investigated. On the surface of all explants yellowish, nodular callus was initiated. The number of explants forming callus depended on the proportion of growth regulators in the induction media. The highest effectiveness of callus formation on media containing Picloram at 25 mcM in combination with BA at 5 or 25 mcM was observed. Nodular callus obtained under influence of BA at concentrations of 5-50 mcM and auxin at higher or the same concentration was capable to differentiation into somatic embryos, whereas nodular callus initiated on media with auxin and lower level of cytokinin (BA at 0.5 mcM) formed only roots
