Rice transformation for resistance to stem borer

Rice transformation for resistance to stem borer was examined by particle bombardment. Embryogenic calli derived from the scutellum of mature seeds of Taipei 309 (japonica), Asemandi, and Rojolele (tropical japonica) were bombarded twice with DNA-coated gold particles. Three plasmids, pUBB [ubiquitin, crylA(b)], pUBC [ubiquitin, crylA(c)], and pSBB [35S, crylA(b)], were used in cotransformation with pRQ6 containing marker genes (gus and hph) or pBargus containing bar and gus genes. The frequency of getting calli resistant to 40 mg L¯¹ hygromycin ranged from 1.8% to 41.4%. A total of 170, 73 and 15 putative transgenic plants (T0) were obtained in Taipei 309, Asemandi, and Rojolele, respectively. The gus expression assay performed on T0 plant-derived calli bombarded with pRQ6/pUBB showed that 66% of Taipei 309 and 25.6% of Asemandi were positive, containing pRQ6. Molecular analysis using PCR and Southern blot, carried out to detect the Bt gene in T1 plants, showed that 25% (6 plants) of Taipei 309 had the crylA(b) gene.
