Diagnosis of YHV infection in Penaeus monodon using monoclonal antibody

Monoclonal antibody specific to yellow head virus (YHV), namely V3-2B was used to detect YHV infection at various time in Penaeus monodon experimentally infected with YHV. Different types of indirect immunoperoxidase assays including dot blot and Western blot were utilized for detection of YHV in haemolymph. Immunocytochemistry was used for detection of YHV infection in haemocyte smears and paraffin sections of various tissues including hepatopancreas, heart, lymphoid organ and gill. The presence of YHV was clearly demonstrated in all tested shrimps with all types of immunoassays after 24 hr of YHV injection. Especially the infection of cells in the haemocyte and all tested tissues were observed before the onset of histopathological symptoms. Therefore, the V3-2B monoclonal antibody can be used to facilitate the accurate diagnosis of early YHV infection.
