Development of random amplified polymorphic DNA (RAPD) technique for identification of pomelo (Citrus grandis)

Pomelo (Citrus grandis) is one of the most popular fruits in Thailand. I was applied random amplified polymorphic DNA (RAPD) technique for identity of the pomelo cultivars. Eighteen cultivars of pomelo have been collected. 144 random primers were screened and 131 primers or about 91 percent of them responded with the pomelo DNA. 30 primers which gave clear amplified products were used to analyze all the DNA samples. The result showed significant differences among 18 cultivars. All cultivars could be identified by using one primer. However, using more primers and simultaneously compared with the standard cultivars should be done for a reliable result. A comparison of RAPD patterns from each cultivar with another can analyzed evolutionary relationship.
