Experimental study of scale-up packed bed column for KOJI fermentation

Study of Soybean fermentation and Aspergillus oryzae in packed bed columns was carried out in two different sizes of packed bed bioreactors. A large bioreactor of 42 liters was scaled up from a small bioreactor of 6.4 liters. Various parameters were measured during 96 hours of fermentation namely protease activity, amylase activity, reducing sugar content, soluble protein content, true protein content, glucosamine content, temperature, moisture content and relative humidity. The results showed that fungi grew slightly better in the small packed bed. Same true protein and enzyme activities were obtained at the end of fermentation of 96 hours. Maximum temperatures in the small and large packed beds were 36 deg C and 42 deg C respectively. The study showed that temperature had dominated effect on the fungal growth. This study pointed out that the scaled up criteria of constant ratio of the air volume/Koji volume and bed height/diameter may not be valid for 6.4 liters to 42 liters. Some adjustment needs to be done to get on appropriate ratio which would directly affect on the bed temperature, protease activity, amylase activity, reducing sugar content, soluble protein content and glucosamine content.
