Development of simultaneous analysis for six penicillins in bovine muscle and kidney using liquid chromatography/mass spectrometry

Using liquid chromatography/mass spectrometry (LC/MS), simultaneous detection was made possible for the analysis of six penicillins in bovine muscle and kidney. The penicillins were extracted using a Macilvain buffer (pH4.0), and the extracted solution was cleaned on a Mega Bond-Elut C18. The extract was dissolved in 2 mmol/l ammonium acetate for injection into the LC/MS. LC separation was performed with a gradient elution of 2 mmol/l ammonium acetate-acetonitrile in the mobile phase. The penicillins were detected by selected ion monitoring using molecular-related ions and fragment ions. The average recoveries of the penicillins from fortified tissues were in the range of 66.7-89.3% in muscles and 67.5-77.9% in kidneys. Detection limits were 0.02microg/g for ampicillin and 0.01microg/g for other penicillins. The present method was used for the analysis of residual ampicillin in kidneys, and the drug was confirmed by the two monitor ions.
