Identification of plaice fillets

英语. An analytical method was developed to distinguish plaice (Pleuronectes platessa) from flounder (Platichthys flesus) fillets. The species Pleuronectes platessa, mainly caught in Northern Europe and in the Atlantic Ocean, and the species Platichthys flesus, which is more frequent in internal seas such as the Mediterranean Sea, are difficult to distinguish when fish is presented in form of cleaned frozen fillets or is pre-cooked and breaded. The method based on the different protein characteristics of the two species differentiated by isoelectrofocusing is not always resolutive. Instead, the analysis of extracted DNA gives results whose interpretation is unequivocal. The restriction of fragment 359 bp amplified from cytochrome b gene presents a fragmentation, evidenced by an electrophoretic run, which, in the case of flounder, is different from the one of plaice

意大利语. E' stato messo a punto un metodo analitico per distinguere filetti di platessa (Pleuronectes platessa) da quelli di passera di mare (Platichthys flesus). La specie Pleuronectes platessa, pescata soprattutto nell'Europa del Nord e nell'Oceano Atlantico, e la specie Platichthys flesus, più frequente nei mari interni come il Mediterraneo, sono difficilmente distinguibili quando il pesce si presenta ripulito sotto forma di filetto congelato o già precotto e impanato. Il metodo che impiega le diverse caratteristiche proteiche delle due specie, differenziate attraverso isoelettrofocalizzazione, non risulta sempre risolutivo. Dall'analisi del DNA estratto, invece, si ottengono dati di sicura interpretazione. La restrizione del frammento 359 bp amplificato dal gene del citocromo b presenta una frammentazione, evidenziata da una corsa elettroforetica, diversa per la passera di mare rispetto alla platessa
