Cultural and molecular characterization of Stenocarpella macrospora (Earle) sutton causing leaf blight, ear rot and stalk rot in maize

Ten isolates of Stenocarpella macrospora (Earle) sutton [syn. Diplodia macrospora (Earl)] causing leaf blight, ear rot and stalk rot on corn, were characterized using different media and substrates. Formation of black pycnidia was observed on oattmeal agar and corn husk three weeks after incubation at 25 degC under continuous artificial light. The optimum mycelial growth of most of the isolates was eithin 25-32 degC. Genomic DNA of the isolates was extracted and quantified. PCR amplification of the internal transcribed spacer (ITS) region of the ribosomal DNA generated an approximately 575 bp band. This study sets the groundwork for S. macrospora reisistance evaluation and to investigate its population structure by PCR technique.
