Production, activity and stability of a novel glucansucrase from a newly isolated strain of Leuconostoc mesenteroides PCSIR AA1

Glucansucrase is an industrially important enzyme produced by Leuconostoc mesenteroides that catalyzes the production of glucan. Glucan is widely used in the pharmaceutical, cosmetic, and food industries. The present study was designed to determine the effects of calcium ions and temperature on enzyme production and its stability. Results showed that glucansucrase production by locally isolated strain Leuconostoc mesenteroides PCSIR AA1 increased up to approximately three folds by the addition of calcium ions (0.01% CaCl2) in the fermentation media as compared to the media without calcium ions. Enzyme production was maximum at 25 degree C in the media containing calcium ions. The extracellular glucansucrase activity increases with the increase in the temperature and after reaching its maxima at 35 degree C the activity declined. Stability data also showed that calcium ions in the culture provide stability to the enzyme for a longer period of time as compared wit h the one without calcium ions, while extracellular glucansucrase is more stable or % loss activity is less when stored at -18 degree C.
