Genetic polymorphism of ACC synthase and ACC oxidase in autochthnous apple cultivars

Allelic polymorphism of the genes encoding ACC synthase and ACC oxidase, enzymes on ethylene biosynthese pathway, was studied in 24 autochthonous apple cultivars from the collection planting of the Fruit and Grape Research Centre in Cacak (Serbia), and compared to their fruit storage capability. Based on the analysis of PCR product of amplified fragment of the gene encoding ACC synthase, two alleles of ACS1 gene were identified: ACS1-1 and ACS1-2, and analyzed autochthonous cultivars were homozygous for allele ACS1-1 or heterozygous (ACS1-1/ACS1-2). Restriction analysis of PCR product of amplified fragment of the gene encoding ACC oxidase in the studied material resulted in the identification of three alleles: a, b and c and four phenotypes: aa, bb, ab and ac. Allele b is a new allele, described for the first time in this paper.
