Study on the number of estrogen and progesterone receptors on mammary gland of bitch with and without tumor mass using immunohistochemical assay

Objectives of this study were to evaluate the correlations between immunohistologically expressions of estrogen receptors(ER-alpha) and progesterone receptors(PR) in histologically normal and tumoral canine mammary tissues and between number of positive cells of both receptors and duration of tumor growth in the same dog. Twelve histologically tumoral mammary tissues and contralateral histologically normal mammary tissues were obtained from 12 dogs by surgical excision. They were fixed in 4 percent paraformaldehyde and embedded in paraffin. Four-micrometers thick sections were cut and stained for the expression of ER-alpha and PR by avidin-biotin-peroxidase complex (ABC) method. The expressions of the ER-alpha and the PR were defined as numbers of immunohistochemically positive cells in the mammary tissues. ER score and PR score were calculated as P1+ (2 X P2) + (3 x P3) where P1, P2 and P3 are the estimated percentages of positive nuclei with low(P1), medium(P2) and high(P3) intensity of immunostaining color. Analysis of the correlations of the expressions of the ER and PR in histologically normal and tumoral mammary tissues, and between ER and PR score and duration of tumor growth within the groups of bitches were made by using Paired T-test and Spearman rank correlation coefficient. The expressions of the ER-alpha and the PR in normal and tumoral mammary tissues were not significantly different (P = 0.17, P = 0.16). A positive correlation was found between the number of the ER-alpha and the PR (r = 0.54,P = 0.005) in both normal and tumoral mammary tissues. The PR expression and duration showed a significant inverse correlation(r = -0.94,P = 0.05). The results from this study are of useful to select the appropriate therapy for canine mammary tumors.
