Preparation of hen egg yolk immunoglobulin (IgY) against recombinant Cholecystokinin-39 of pig

中国人. 制备重组猪三十九肽胆囊收缩素（GST-CCK39），以其为抗原免疫28周龄的蛋鸡，用水稀释脱脂处理，聚乙二醇（PEG）一步法制备高效价卵黄抗体，并用免疫琼脂扩散试验、ELISA、Dot-ELISA检测其效价及特异性。结果表明，免疫琼脂扩散测得特异性抗体效价达1∶16以上，ELISA 效价达1∶12 800以上；Dot-ELISA显示所获得卵黄抗体具有较强的特异性；高效价，特异性的卵黄抗体可维持50 d以上。表明用重组蛋白GST-CCK39免疫蛋鸡后能获得效价高、特异性强的卵黄抗体。
显示更多 [+] 显示较少 [-]英语. In order to prepare the immunoglobulin Y for the fusion protein GST-CCK39, hens were immunized with partially purified fusion protein GST-CCK39. One-step polyethylene glycol (PEG) precipitation procedure was used to purify the immunoglobulin Y (IgY). The titers of purified IgY were detected by Agar Gel Precipitation Text (AGPT), Enzyme-linked Immunosorbent Assay (ELISA) and Dot-ELISA. Results showed that the AGPT titer in the IgY was higher than 1∶16; the ELISA titers in the purified IgY was higher than 1∶12 800. High titers IgY to GST-CCK39 could be induced by immunization successfully, and the specific activity could last more than 50 days. All these indicated that the good specificity and high titers hen egg yolk immunoglobulin ( IgY) could be prepared by fusion protein GST-CCK39.
