Improvement of protein secretion capability of Saccharomyces cerevisiae DBY 746 employing chemical mutagenesis

Saccharomyces cerevisiae DBY 746 was subjected to chemical mutagenesis to obtain supersecretion mutants with elevated levels of protein secretion into the culture supernatant. Mutagenesis was carried out by using ethyl methane sulphonate (EMS). A number of mutants (20-30 colonies) obtained were screened based on their capability in secreting invertase into the culture supernatant. Protein secretion capability was also analysed by measuring invertase secreted into the culture supernatant following growth of the cell in sucrose-containing medium. It was observed that at 2 percent sucrose concentration, mutagenesis treatment increased level of protein secretion into the culture supernatant. The level of invertase secreted was also induced by the increased concentration of sucrose used as the carbon source. However, M15 mutant strain showed lower capacity of protein secretion than the parental strain under 6 percent sucrose concentration.
