Polymorphism at K-casein and prolactin loci in goats investigatyed by pcr-rflp and dna sequncing.

The DNA sequence variations at the K-casein(K-CN) and prolactin (PRL) genes were investigated in Malabari, Attappady black, Jamnapari and Salem black goats by PCR-RFLP and DNA sequencing methods. A 0.9 kb region of the K-CN gene enclosing the polmorphic Rsal site within the third exon were amplified separately byPCR. In the case of K-CN locus the digestion of PCR products with restriction enzymes Hind III and Rsal revealed fragment pattern characteristic of the B allele in all the animals tested. The digestion of the PCR product from PRL locus with Rsal enzyme revealed a single allele, namely, the allele A (156 bp) in all the breeds tested. Following sequencing, the PCR product was found to be of 156 bp in length and the BLAST search at the NCBI site revealed 100; sequence homology with corresponding sequence of the ovine mammary gland prolactin mRNA (Assession X76050.1), caprine pituitary gland prolactin mRNA (Accession X76049.1) and caprine mammary gland prolactin mRNA (Accession X76048.1). The sequences were submitted to GenBank and obtained accession numbers AY517552 for Malabari and AY534756 for Jamnapari, respectively.
